Troopers said a road rage incident between the two drivers sparked the encounter.
When the semi driver came up alongside the box truck, the driver in the semi started shooting, hitting the passenger side of the truck, but missing the 42-year-old driver, troopers said.
The box truck driver swerved left into the shoulder away from the gunfire and stopped for help, while the semi driver continued north on I-95, FHP said.
Troopers said witnesses were able to describe the semi, which was broadcast to other law enforcement agencies and used to contact the company that owned the vehicle.
The company told troopers where the semi might be, and Georgia State Patrol used the info to track down the tractor-trailer on I-95 in Georgia.
The driver was arrested and charged with aggravated assault with a deadly weapon, FHP said.
Troopers said the incident is still under investigation, and anyone with information about what happened is asked to contact FHP investigators at 904-301-3700.
